Effects of Soybean Isoflavone Genistein and Daidzein on Proliferation and Viability of Human Osteogenic Sarcoma Cell Saos-2

Abstract
Backglound: Soybeans abundantly contain genistein and daidzein in isoflavones (see in figures). Difference of its structure is with or without hydroxyl group at 5-position. It has been reported that these isoflavones have anti-oxidant and anti-cancer effects. In this study, we investigated effects of genistein and daidzein on proliferation and viability of human osteogenic sarcoma cell Saos-2.
Methods: Saos-2 cells were gifted from Riken Cell Bank (Tsukuba, Japan). Genistein and daidzein were purchased from Tokyo Kasei Co., Japan. We monitored the proliferation of Saos-2 cells treated with 0 to 100 μM genistein and daidzein under a microscope for 5 days. We also examined viability of Saos-2 cells, at 48 h and 72 h, treated with 0 to 100 μM genistein and daidzein by using Cell Counting kit-8 (Dojindo).
Results: Genistein significantly inhibited proliferation and viability of Saos-2 cells in a dose-dependent manner. At 100 μM genistein, viability of Saos-2 cells was reduced by 50%. By contrast, daidzein weakly inhibited proliferation and viability of Saos-2 cells. Conclusions: We found that 5-position hydroxyl group of genistein play a critical role on inhibitory effects of proliferation and viability of Saos-2 cells.
